I think that just means that you'll need to sterilize everything that touches the microbes. I like how the teacher above me wrote that you need to use 'good english'...
Plus, you don't autoclave plates ready for innoculation... You autoclave the medium, pour your plates, stabs, or slants, then innoculate them. After you're done, you'll probably have to autoclave everything again to kill the microbes.

You firstly need to decide on one factor to investigate, eg temperature, pH, etc, and then keep all other variables constant.

Then as its A2 coursework you wil have to do your own backgroud reading and research (I know its boring! Ive just done mine!!). Research into the enzymes that are used in alcohol metabolism. Then once youve found the enzyme, your school science lab should be able to provide you with it, or tell you where to get hold of some that can be used experimentally.

Then you need to work out what you are going to measure - either rate of production of product, or rate of use of reactant, and decide how this can be differentiated. Eg, is there any indicator you could usethat will change colour when the reaction is complete....again this will require your own research. Then simply time how long it takes for the reaction to be completed under the varying factor. Eg if investigating temperature, do the reaction at 30, 40, and 50 degreesC. Remember to replicate the whole experiment about 10 times!!

Good luck....you seem to have chosen a very difficult topic here.....i did something a bit simpler which looked at the effect of the presence of bile in lipid hydrolysis....this was much easier to carry out as the reactants were available readily in the lab (Milk, bile salts, and lipase enzyme), and an indicator could be used too. But if you want to go with what you are interested in...the alcohol thing...then good for you!! go for it!!
